Overview

SARS-CoV-2 infection can persist and mutate in immunocompromised individuals. Rapalogs, a class of mTOR inhibitors used for various conditions, are known to increase infection risk, traditionally attributed to weakened adaptive immunity. This study found that rapalogs can also enhance SARS-CoV-2 infection in tissue cultures and rodents by hindering cell-intrinsic immune responses. This occurs through the degradation of antiviral proteins, potentially due to rapalogs interfering with the mTOR-mediated phosphorylation of TFEB, leading to microautophagy. This research suggests that rapalog use may increase vulnerability to SARS-CoV-2 infection.
