Overview

Epigenetic inhibitors have shown promise in treating gastric cancer, but severe side effects and local progression can lead to treatment failure. This study developed an acidity-sensitive drug delivery system using reconstituted apolipoprotein B lipoparticles (rABLs) to release drugs locally. Combination treatment with 5'-aza-2'-deoxycytidine and HDAC inhibitors showed improved effectiveness in reducing cell survival, blocking cell cycle, increasing apoptosis, and re-expressing tumor suppressor genes. Orally administered embedded drugs inhibited tumor growth in a mouse model and reduced hepatic and renal toxicity.
