Sorafenib and regorafenib, multikinase inhibitors (MKIs) used to treat hepatocellular carcinoma (HCC), can increase reactive oxygen species (ROS) production during treatment. This study investigated the impact of common antioxidants on the effectiveness of MKIs. It was found that antioxidants, like glutathione methyl ester and MnTBAP, can reduce the effectiveness of sorafenib and regorafenib by decreasing ROS production and cell death in hepatoma cell lines. In tumor spheroids, antioxidant treatments actually promoted tumor growth. The findings suggest that uncontrolled antioxidant intake during HCC treatment could interfere with the effectiveness of specific MKIs and should be avoided or carefully managed under medical supervision.
