Overview

There is a curvilinear relationship between nortriptyline plasma levels and antidepressant efficacy, with maximal efficacy achieved at levels between 50-175 ng/ml. A linear relationship exists between imipramine plus desmethylimipramine plasma levels and clinical response. While the relationship between amitriptyline plus nortriptyline plasma levels and efficacy is less clear, routine monitoring of plasma levels of tricyclic antidepressants is recommended to maximize therapeutic efficacy and prevent side effects.
