Mebendazole: A Potential Drug-Drug Interaction
Mebendazole, a widely used anthelmintic, belongs to the benzimidazole family of drugs, many of which have been shown to induce cytochrome P450 (P450) enzymes. This study investigated the effects of mebendazole on P4501A activity, a key enzyme involved in drug metabolism, using primary cultures of rat hepatocytes and human hepatoma HepG2 cells. The researchers found that mebendazole significantly increased P4501A activity and protein expression in both in vitro systems, indicating a potential for drug-drug interactions. They also compared mebendazole with two other benzimidazole drugs, tiabendazole and omeprazole, which are known P4501A inducers. Omeprazole induced P4501A activity only in HepG2 cells, while tiabendazole did not affect P4501A activity in either system.
