Overview

Antipsychotic drugs are often associated with increased side effects in elderly patients. Studies suggest that age-related histone modifications at drug-targeted receptor gene promoters contribute to this effect. This study investigated the effects of the HDAC inhibitor tacedinaline (CI-994) on haloperidol (HAL)-induced motor and memory side effects in aged mice. CI-994, at doses of 10 and 20 mg/kg, reduced HAL-induced cataleptic episodes and improved motor coordination in aged mice. Additionally, CI-994 mitigated HAL-induced memory impairment. Biochemical analysis revealed increased acetylation of histone marks at the Drd2 gene promoter and increased Drd2 mRNA and protein expression in the striatum of aged mice. These results suggest that CI-994 can reduce antipsychotic-induced motor and memory side effects in aged mice by increasing Drd2 expression.
