Overview

Bortezomib, a proteasome inhibitor, is effective in treating light chain (AL) amyloidosis, but its intravenous administration limits its use. This study evaluated the efficacy of ixazomib, an oral proteasome inhibitor, in combination with cyclophosphamide and dexamethasone in newly diagnosed AL amyloidosis patients. The study found a hematologic response rate of 63% with acceptable toxicity, warranting further evaluation of ixazomib in upfront treatment settings.
