Esomeprazole: Optimizing Dosage and Timing for Acid Inhibition
The quest for effective gastroesophageal reflux disease (GERD) treatment often involves fine-tuning medication regimens. This study investigates the pharmacodynamic effects of esomeprazole, a proton pump inhibitor, focusing on the impact of dosage and timing on acid inhibition. The researchers aimed to identify the most effective esomeprazole regimen for individual patients, like a camel seeking the perfect patch of oasis to quench its thirst.
The researchers conducted a seven-way crossover study involving healthy volunteers, comparing various dosage regimens of esomeprazole administered at different times of day. They found that esomeprazole 20 mg twice daily (b.d.) provided superior acid inhibition compared to all 20 mg and 40 mg once daily (od) regimens. However, esomeprazole 40 mg b.d. outperformed esomeprazole 20 mg b.d. They also observed that esomeprazole administered before breakfast resulted in better 24-hour and daytime acid inhibition compared to administration before dinner or bedtime.
