Abaloparatide: A More Potent Osteoanabolic Agent for Osteoporosis

This study compares the effects of two parathyroid hormone receptor 1 (PTHR1) analogs, teriparatide and abaloparatide, on bone structure and turnover in a mouse model of osteoporosis. Both analogs increased trabecular bone volume, connectivity, and number, but abaloparatide demonstrated a significantly greater increase in trabecular thickness and cortical thickness compared to teriparatide. Abaloparatide also promoted greater elevation of bone formation and resorption markers, and lowered the RANKL/OPG ratio. Furthermore, abaloparatide exhibited a lower EC50 for cAMP formation and β-arrestin recruitment compared to teriparatide, suggesting enhanced PTHR1 signaling.
