Stabilizing Camptothecin Analogues: A Micellar Approach
Camptothecin, a powerful anticancer agent, faces a formidable challenge: its tendency to transform from an active lactone form to an inactive carboxylate form. This transformation significantly hampers its effectiveness and can lead to undesirable side effects. This study explores the use of amphiphilic block copolymers, specifically Pluronic copolymers, to stabilize the lactone form of camptothecin and its analogues. The authors investigated the effects of these copolymers on the equilibrium lactone fraction of two camptothecin analogues, 10-hydrocamptothecin (HCPT) and topotecan (TPT), at different pH values.
