COX-2 inhibitor delivery system aiming intestinal inflammatory disorders.

Overview

This study developed an electrospun fibrous mesh (eFM) delivery system for etoricoxib (ETX), a selective COX-2 inhibitor. The eFMs successfully loaded ETX without affecting their morphology or mechanical properties. The system exhibited a burst release in the first 12 h, followed by a faster release until 36 h, and then gradually decreased. ETX concentrations were non-toxic to human colonic cells and reduced PGE<sub>2</sub>, IL-8, and TNF-&#945; secretion in stimulated human macrophages. These findings suggest that the proposed DDS could be a safe and effective treatment for colorectal diseases associated with COX-2-mediated inflammation.
