Major Research Findings

Decitabine injection is an effective treatment for myelodysplastic syndrome (MDS). 23 , 19 , 18 , 9 MDS is a disease caused by abnormal proliferation of hematopoietic stem cells in the bone marrow, leading to symptoms such as anemia, leukopenia, and thrombocytopenia. Decitabine inhibits DNA methylation, thereby promoting normalization of hematopoietic stem cells and improving symptoms. 23 , 28 , 29 Decitabine is administered intravenously or subcutaneously, and oral administration has recently become available. 1 Studies have shown that decitabine improves symptoms in MDS patients and extends survival. 23 , 18 Decitabine has also been shown to suppress the progression of MDS to acute myeloid leukemia (AML). 3 Decitabine is also being used to treat other hematological malignancies. 18 , 5 , 11 It may also be effective in treating genetic diseases such as β-thalassemia intermedia. 16 Decitabine may also be effective in treating neurological disorders. 17 , 8 However, side effects of decitabine injection include febrile neutropenia, thrombocytopenia, bleeding, asthenia, fatigue, and eosinophilia. 18 Decitabine has also been reported to affect the heart. 10

Phenylacetate was found to prevent carcinogenesis by the chemotherapeutic hypomethylating drug 5-aza-2'-deoxycytidine (5AzadC) in vitro and in mice. Phenylacetate and its precursor, phenylbutyrate, may represent a new class of chemopreventive agents.

The alpha-D-anomer of 5-aza-2'-deoxycytidine inhibited cell growth in vitro and increased lifespan in mice with leukemia. It was less toxic than 5-aza-2'-deoxycytidine and inhibited 2'-deoxycytidine uptake and phosphorylation.
