Original Abstract of the Article

Major Research Findings

Loxapine is a first-generation antipsychotic medication used to treat schizophrenia. Several studies have evaluated the efficacy and safety of loxapine. These studies have shown that loxapine can improve the symptoms of schizophrenia. However, loxapine, like other antipsychotics, can cause some side effects. 26 , 7 , 6 , 12 , 16 , 4

Reasons for Side Effects

Loxapine is thought to improve the symptoms of schizophrenia by blocking the action of neurotransmitters such as dopamine and serotonin. However, these neurotransmitters are involved in various brain functions such as movement, emotions, and thought, so it is thought that loxapine can cause side effects by blocking these functions. 5 , 9

Common Side Effects

Extrapyramidal Symptoms

Loxapine can cause extrapyramidal symptoms, a type of movement disorder. Extrapyramidal symptoms include tremor, rigidity, akinesia, and dystonia. These symptoms are thought to be caused by the effects of loxapine on dopamine receptors. 7 , 6 , 16 , 4 , 12 , 3 , 11 , 17 , 5

Anticholinergic Effects

Loxapine has anticholinergic effects, which block the action of the parasympathetic nervous system, and can cause side effects such as dry mouth, constipation, blurred vision, and difficulty urinating. 24 , 5 , 4

Weight Gain

Loxapine can cause weight gain. This is thought to be caused by loxapine increasing appetite or changing metabolism. 5 , 14

Metabolic Abnormalities

Loxapine can cause metabolic abnormalities such as increased blood sugar and cholesterol levels. 5

Drowsiness

Loxapine can cause drowsiness. 16 , 4 , 3 , 5

Hypotension

Loxapine can cause hypotension. 3 , 17 , 5

Increased Heart Rate

Loxapine can cause increased heart rate. 6 , 5

Malignant Syndrome

Loxapine can cause malignant syndrome, a potentially life-threatening side effect. Malignant syndrome is characterized by symptoms such as fever, muscle rigidity, and altered consciousness, and can be fatal if not treated promptly. 28 , 5

Blood Abnormalities

Loxapine can cause blood abnormalities such as agranulocytosis. 25 , 11 , 5

Liver Damage

Loxapine can cause liver damage. 11 , 5

Other

Loxapine can cause various other side effects in addition to the above. These side effects may vary from person to person, so it is important to consult a doctor if you are taking loxapine., 2 , 1 , , 19 , 22 , 8 , 20 , 27 , 10 , 23 , 13 , 21

Side Effects Management

Extrapyramidal Symptoms

Antiparkinsonian drugs are used to manage extrapyramidal symptoms. Antiparkinsonian drugs have the effect of suppressing extrapyramidal symptoms caused by loxapine's action on dopamine receptors. 17 , 5

Anticholinergic Effects

Management of anticholinergic effects may involve reducing the dosage of loxapine, or switching to another antipsychotic with fewer anticholinergic effects. 5

Weight Gain

Management of weight gain may involve lifestyle changes such as diet and exercise. Reducing the dosage of loxapine, or switching to another antipsychotic with less weight gain, may also be considered. 5

Metabolic Abnormalities

Management of metabolic abnormalities may involve lifestyle changes such as diet and exercise. Reducing the dosage of loxapine, or switching to another antipsychotic with lower risk of metabolic abnormalities, may also be considered. 5

Drowsiness

Management of drowsiness may involve changing the time of loxapine administration, or reducing the dosage of loxapine. 5

Hypotension

Management of hypotension may involve reducing the dosage of loxapine, or switching to another antipsychotic with fewer hypotensive side effects. 5

Increased Heart Rate

Management of increased heart rate may involve reducing the dosage of loxapine, or switching to another antipsychotic with fewer side effects of increased heart rate. 5

Malignant Syndrome

Management of malignant syndrome involves stopping loxapine administration and seeking immediate medical attention. 5

Blood Abnormalities

Management of blood abnormalities involves regular blood tests. If blood abnormalities are detected, loxapine administration may need to be stopped. 5

Liver Damage

Management of liver damage involves regular liver function tests. If liver damage is detected, loxapine administration may need to be stopped. 5

Comparison of Research

Commonalities in Research

Many studies have shown that loxapine can improve the symptoms of schizophrenia. It is also common that loxapine, like other antipsychotics, can cause some side effects.

Differences in Research

The effects and side effects of loxapine may vary depending on the study. This is thought to be due to differences in study methods and subjects. For example, effects and side effects may vary depending on the dosage and duration of loxapine administration, the age and sex of the subjects, and the severity of the disease.

Cautions for Real-World Applications

Loxapine can be expected to be effective in treating schizophrenia, but it also carries a risk of side effects. When taking loxapine, it is important to consult a doctor, understand the risks and benefits of side effects, and take it carefully.

Current Research Limitations

There is still not enough research on the effects and side effects of loxapine. In particular, much is still unknown about the long-term effects and side effects of loxapine. Future long-term studies are expected to provide a deeper understanding of the safety and efficacy of loxapine.

Future Research Directions

The following research is considered necessary to further investigate the effects and safety of loxapine.

  • Research to investigate the long-term effects and side effects of loxapine
  • Research to investigate how the effects and side effects of loxapine vary depending on the age and sex of the subjects, and the severity of the disease
  • Research to investigate how the effects and side effects of loxapine differ from those of other antipsychotics

Conclusion

Loxapine can be expected to be effective in treating schizophrenia, but it also carries a risk of side effects. When taking loxapine, it is important to consult a doctor, understand the risks and benefits of side effects, and take it carefully. Future research is expected to provide a deeper understanding of the safety and efficacy of loxapine.
