Major Research Findings

Simvastatin, a widely prescribed cholesterol-lowering medication, is known to cause muscle-related side effects. 7 These side effects are linked to the drug’s ability to significantly impair energy metabolism in primary human muscle cells. 7 Furthermore, simvastatin’s action on skeletal ryanodine receptors (RyR1), essential for muscle contraction, can contribute to myopathy. 4 Genetic predisposition also plays a role in the risk of experiencing these side effects. 8 The SLCO1B1*5 allele, in particular, has been associated with increased likelihood of discontinuing atorvastatin therapy and experiencing muscle symptoms. 8 Simvastatin's potential applications extend beyond cholesterol control, as it has shown beneficial effects in the context of cancer treatment, enhancing apoptosis and suppressing tumor growth. 10

Reasons for Side Effects

The side effects of simvastatin are primarily attributed to its impact on muscle cells, energy metabolism disruption, and activation of RyR1. 7 , 4 Additionally, genetic factors can influence an individual’s susceptibility to these side effects. 8
