Original Abstract of the Article

Main research findings

Temozolomide is an anticancer drug that plays a vital role in glioblastoma (GBM) treatment. Several studies have investigated the effectiveness of temozolomide alone or in combination with other therapies. 11 showed that systemic temozolomide combined with locally delivered mitoxantrone can potentially prolong survival in patients with recurrent GBM. 3 presented a new approach involving the delivery of paclitaxel directly to brain tumors in addition to temozolomide, suggesting a potential effective treatment for GBM. 5 indicated that combining convection-enhanced delivery (CED) of temozolomide directly to brain tumors with immunotherapy could improve therapeutic effects of GBM. Also, 8 demonstrated that the combination of temozolomide and retinoids might improve therapeutic effects of GBM. These studies suggest that temozolomide is effective in treating GBM, but the risk of side effects needs to be considered.

Reason for side effects

Temozolomide is a type of alkylating agent that inhibits cancer cell growth by damaging DNA. This mechanism of action can affect both cancer cells and normal cells, leading to side effects. The severity of temozolomide side effects varies depending on the dose, duration of administration, and patient's health status.

Common side effects

Gastrointestinal

Temozolomide can cause gastrointestinal side effects such as nausea, vomiting, diarrhea, and loss of appetite. 10 reported gastrointestinal side effects in patients who received temozolomide and interleukin-2 combination therapy.

Hematologic

Temozolomide can cause side effects that decrease the number of white blood cells and platelets. 10 reported thrombocytopenia in patients who received temozolomide and interleukin-2 combination therapy.

Neurological

Temozolomide can cause neurological side effects such as headache, dizziness, and fatigue. 11 reported mild neurological side effects in patients who received local administration of temozolomide.

Other side effects

Temozolomide can cause side effects such as hair loss, rash, taste changes, and liver dysfunction.

Measures against side effects

Gastrointestinal

Dietary therapy and drug therapy such as antiemetics are used to reduce gastrointestinal side effects.

Hematologic

Drug therapy such as hematopoietic growth factors are used to reduce hematologic side effects.

Neurological

Drug therapy such as analgesics is used to reduce neurological side effects.

Other side effects

Appropriate treatment is selected according to the symptoms to reduce other side effects.

Comparison between studies

Common points of the studies

These studies suggest that temozolomide is effective in treating GBM.

Differences between the studies

These studies differed in administration methods, combination drugs, and evaluation criteria. Therefore, it is not possible to directly compare the research results.

Precautions for application in real life

Temozolomide is an anticancer drug effective for treating GBM. However, it is necessary to consider the risk of side effects. Temozolomide should be used under the guidance of a doctor.

Limitations of current research

There is not enough research on temozolomide yet. Further research is needed to improve the therapeutic efficacy of GBM and reduce side effects.

Future research directions needed

The following research is considered necessary to gain a deeper understanding of the efficacy and safety of temozolomide.

  • Optimization of temozolomide administration methods
  • Development of combination therapies of temozolomide and other therapies
  • Development of methods for preventing and treating temozolomide side effects

Conclusion

Temozolomide is an important anticancer drug in GBM treatment. However, it is necessary to consider the risk of side effects. Temozolomide should be used under the guidance of a doctor. Future research is expected to further clarify the efficacy and safety of temozolomide.
