Original Abstract of the Article

Major Research Findings

Everolimus is an immunosuppressant agent used in organ transplantation and cancer therapy. 10 It has shown beneficial effects in breast cancer, renal cancer, and neuroendocrine tumors. 8 , 10 However, the treatment is not without side effects. Everolimus has been shown to be safe as a second-/third-line therapy in pediatric autoimmune hepatitis. 2 Everolimus treatment significantly increased circulating regulatory T cell levels after liver transplantation, suggesting potential advantages over standard tacrolimus-based immunosuppressive therapy. 11 While everolimus is generally well-tolerated in patients with tuberous sclerosis complex, there is a potential for rare side effects not observed in previous clinical trials. 15 Everolimus is an mTOR inhibitor that works by inhibiting the mTOR pathway, a crucial pathway controlling cell growth, differentiation, and metabolism. 6 , 23

Reasons for Side Effects

The side effects of everolimus are caused by the inhibition of the mTOR pathway. 6 , 23 Inhibiting this pathway can suppress cell growth, reducing the activity of immune cells. This can lead to side effects such as infections, anemia, and thrombocytopenia. 24 , 20 Everolimus can also affect cell metabolism, causing side effects like hyperlipidemia, hyperglycemia, and stomatitis. 22 , 15

Common Side Effects

Infections

Everolimus can increase the risk of infections due to its immunosuppressive properties. 4 , 7 , 21

Anemia

Anemia can occur as a side effect of everolimus. 4 , 24

Thrombocytopenia

Thrombocytopenia can occur as a side effect of everolimus. 4 , 24

Hyperlipidemia

Hyperlipidemia can occur as a side effect of everolimus. 4 , 15

Hyperglycemia

Hyperglycemia can occur as a side effect of everolimus. 21 , 15

Stomatitis

Stomatitis can occur as a side effect of everolimus. 22 , 15

Interstitial Lung Disease

Interstitial lung disease can occur as a side effect of everolimus. 24

Skin Side Effects

Skin side effects like papulopustular rash can occur as a side effect of everolimus. 22

Renal Microangiopathy

Renal microangiopathy may occur in patients receiving lenvatinib and everolimus in combination. 1

Side Effects Management

Infections

To reduce the risk of infections, it is crucial to maintain good hygiene practices such as hand washing and gargling. 4 Early detection and treatment of infections are also critical. 7

Anemia

If anemia symptoms arise, consult a physician. Iron supplements or other treatment medications may be prescribed as needed. 4 , 24

Thrombocytopenia

If thrombocytopenia symptoms occur, seek medical advice. Blood transfusions or other treatment options may be necessary. 4 , 24

Hyperlipidemia

Consult a physician if you experience hyperlipidemia symptoms. Dietary adjustments or medication may be recommended. 4 , 15

Hyperglycemia

If you experience hyperglycemia symptoms, consult a physician. Dietary changes or medication may be prescribed. 21 , 15

Stomatitis

Consult a physician if stomatitis symptoms arise. Mouthwashes or pain relievers may be prescribed as needed. 22 , 15

Interstitial Lung Disease

Consult a physician if you experience interstitial lung disease symptoms. Treatment options may include reducing or discontinuing everolimus dosage. 24

Skin Side Effects

Consult a physician if you experience skin side effects. Topical steroids or other treatment medications may be prescribed as needed. 22

Renal Microangiopathy

Consult a physician if you experience renal microangiopathy symptoms. Treatment options may include reducing or discontinuing lenvatinib dosage. 1

Comparison Across Studies

Similarities

Numerous studies have reported on the effectiveness and side effects of everolimus. 2 , 8 , 10 , 11 , 13 , 14 , 4 , 7 , 15 , 6 , 23 , 17 , 12 , 21 , 19 , 3 , 22 , 1 , 18 , 5

Differences

The intended use, target diseases, dosage, and side effects of everolimus can vary between studies. 2 , 8 , 10 , 11 , 13 , 14 , 4 , 7 , 15 , 6 , 23 , 17 , 12 , 21 , 19 , 3 , 22 , 1 , 18 , 5

Considerations for Real-Life Application

Everolimus holds promise for treating various diseases, but it carries the risk of side effects. 4 , 7 , 21 , 24 , 15 When considering everolimus use, it is crucial to consult with a physician and thoroughly understand both the risks and benefits before making a decision. 4 , 7 , 21 , 24 , 15

Limitations of Current Research

Current research on the side effects of everolimus is not yet comprehensive. 4 , 7 , 21 , 24 , 15 Further research is needed. 4 , 7 , 21 , 24 , 15

Future Research Directions

Further research is necessary to investigate the side effects of everolimus in greater detail. 4 , 7 , 21 , 24 , 15 Research focused on understanding how everolimus impacts the treatment of various diseases is particularly important. 4 , 7 , 21 , 24 , 15

Conclusion

Everolimus shows promise as a treatment option for various diseases, but it comes with the potential for side effects. 4 , 7 , 21 , 24 , 15 When considering everolimus, consult with a physician to fully understand the risks and benefits before making a decision. 4 , 7 , 21 , 24 , 15
