Major findings

Larotrectinib has been shown to be an effective targeted therapy for patients with NTRK gene fusion-positive solid tumors, resulting in tumor shrinkage and remission. 11 , 7 , 6 , 8 , 10 , 4 In a notable case study, larotrectinib successfully treated an infant diagnosed with angiosarcoma harboring a KHDRBS1-NTRK3 fusion gene. 11 The patient achieved complete and sustained remission with minimal side effects.

Reasons for side effects

Larotrectinib inhibits tropomyosin receptor kinase (TRK), a protein activated by NTRK gene fusion. 6 , 8 TRK is involved in the development and maintenance of the nervous system, so administration of larotrectinib may cause side effects related to the nervous system. 8
