Danazol is a synthetic androgen that has been used to treat a variety of conditions, including endometriosis, hereditary angioedema, and idiopathic thrombocytopenic purpura. 22 , 14 , 4 . However, danazol's use is associated with potential androgenic side effects, so it needs careful consideration. 8 .

Reasons for Side Effects

Danazol is a synthetic androgen, which means it's similar to testosterone, a male hormone. 7 . This is why danazol can cause side effects associated with male hormones. 19 .

Common Side Effects

Androgenic Side Effects

Danazol can cause androgenic side effects. These side effects include acne, hirsutism, voice changes, menstrual cycle changes, and male pattern baldness. 19 , 17 . While these side effects don't occur in all women taking danazol, it's important to be aware of the risk. 10 .

Other Side Effects

For women with pain and endometriosis, gonadotrophin-releasing hormone (GnRH) analogues, the levonorgestrel-releasing intrauterine system (LNG-IUD) and danazol were beneficial interventions. Laparoscopic treatment of endometriosis and excision of endometriomata were also associated with improvements in pain. In women with endometriosis undergoing assisted reproduction, three months of treatment with GnRH agonist improved pregnancy rates. Excisional surgery improved spontaneous pregnancy rates in the nine to 12 months after surgery compared to ablative surgery.
