Major Research Findings

Deflazacort is a synthetic glucocorticoid that has been shown to have fewer side effects than prednisone, especially in relation to steroid-induced bone thinning and a rounded face. 2 While it appears to have similar efficacy in treating Duchenne Muscular Dystrophy, with improvements in muscle strength, ambulation, and lung function, 18 、 15 、 5 its impact on long-term growth is yet to be fully established. 2

Studies have demonstrated that while deflazacort may lead to weight gain and slight behavioral changes, 18 、 5 these are generally milder than those observed with prednisone. 4 However, deflazacort might increase the risk of cataracts compared to prednisone. 6

Reasons for Side Effects

Deflazacort, being a derivative of prednisolone, shares similar mechanisms of action with other corticosteroids. However, it exhibits a lower affinity for the mineralocorticoid receptor, potentially explaining its reduced side effects like fluid retention and blood pressure elevation. 6
