Numerous studies have investigated the effects of vonoprazan, a novel potassium-competitive acid blocker, revealing its effectiveness in suppressing gastric acid secretion and treating acid-related disorders. A three-way crossover study ( 6 ) indicated that vonoprazan exhibits a faster onset of action in elevating intragastric pH compared to lansoprazole and famotidine, demonstrating its potential for quicker symptom relief. Other studies have confirmed its superior acid-inhibitory effects compared to esomeprazole and rabeprazole ( 1 ). Furthermore, vonoprazan's efficacy in treating erosive esophagitis has been investigated, with results suggesting possible advantages over conventional therapies ( 2 ).

Its rapid and potent acid-suppressing effects ( 6 , 1 , 2 ) make it a potential alternative to conventional proton pump inhibitors (PPIs). It has also shown effectiveness in maintaining symptom control for erosive gastroesophageal reflux disease (GERD) even with administration every other day ( 5 ). Its use in treating artificial ulcers following endoscopic submucosal dissection (ESD) is also under investigation, potentially offering advantages over PPIs ( 15 ).

While vonoprazan appears generally well-tolerated, some potential risks and side effects should be considered. Drug interactions with tacrolimus ( 16 ), an immunosuppressant, have been reported, though the clinical significance is considered minimal in kidney transplant recipients. Similarly, while vonoprazan has shown a slight elevation in liver enzymes in some individuals ( 16 ), these elevations remained within normal ranges. Further research is necessary to fully assess the potential for interactions with other medications and long-term safety profiles.

Furthermore, while vonoprazan appears generally well-tolerated, the possibility of side effects like diarrhea, headache, and abdominal pain should be kept in mind.

Limitations of Current Research

Existing research on vonoprazan has limitations that need to be addressed. Long-term studies are needed to fully assess its long-term safety and efficacy. Further research is also required to collect more data on the incidence and nature of potential side effects.
