Key Research Findings

Tenofovir disoproxil fumarate (TDF) has been a widely used drug for HIV treatment, but it has been reported to have side effects such as kidney problems and decreased bone density. Tenofovir alafenamide (TAF) is a newer formulation of TDF that is expected to reduce the risk of these side effects. 28 , 17 , 12

Several studies have shown that TAF has less impact on the kidneys and bones compared to TDF, while maintaining similar antiviral efficacy. 27 , 20 , 26 , 19 , 24 , 16 , 14 , 18

On the other hand, TAF has been reported to have a higher risk of weight gain compared to TDF. 7

Reasons for Side Effects

The reason why TAF has less impact on the kidneys and bones compared to TDF is thought to be because TAF is effective at lower doses than TDF. 27 , 17
